Action of spleen exonuclease on transfer ribonucleic acid.

چکیده
The hydrolytic action of spleen exonuclease on serine transfer RNA has been investigated. It was found that two different mechanisms of attack obtain: (a) processive degradation and (b) random cleavage. These two diiferent modes of action are determined by the presence or the absence of the terminal phosphate group at the 5’ terminus of transfer RNA. The loss of several important biological activities of transfer RNA or aminoacyl transfer RNA, such as amino acid acceptor activity, AMP acceptor activity, polynucleotidedirected ribosomal binding, and incorporation of the amino acid moiety into growing polypeptide chains, were examined in detail as a function of the digestion by spleen exonuclease.

Action of Spleen Phosphodiesterase upon Yeast Soluble Ribonucleic Acid.
A major concern in attempts to relate the structure of soluble ribonucleic acid with biological specificity is whether an entire molecule of the acid is needed for biological function. Specific monophosphatases and esonucleases offer a tool with which to approach this question. Hydrolysis of S-RNA’ with venom phosphodiesterase (1) destroys the amino acid-accepting ability of S-RNAs (2, 3), pres...

Amber (am) mutants of the two closely linked sites, B22 and C125, in bacteriophage T4 gene 43 [deoxyribonucleic acid (DNA) polymerase] synthesize in the nonpermissive (su(-)) Escherichia coli host gene 43 products which are devoid of DNA polymerase activity, but which retain a 3'-exonuclease activity. Diethylaminoethyl-cellulose chromatographic analysis of DNA polymerase and deoxyribonuclease a...

Ribonuclease II action on polyuridylate is competitively inhibited by transfer ribonucleic acid and noncompetitively inhibited by sodium ions. At low substrate levels, adenosine-5'-triphosphate is also inhibitory.

The preparation of synthetic ribonucleic acid in which one of the nucleotide species is labeled with P32, makes possible studies on the distribution of the labeled nucleotide in the polynucleotide chain. Fig. 1 presents the structure of a polynucleotide in which labeled adenylic acid is randomly distributed. Hydrolysis of such a polymer with snake venom phosphodiesterase (Fig. 1A) will yield a ...

In the cell-free inductor demonstrated by Niu & Twitty (1953) the effective material was later characterized by Niu (1956) as ribonucleoprotein with the suggestion that the ribonucleic acid (RNA) fraction was the active component. Subsequently Niu (1958a, b) reported tissue-specific (thymus-like) induction in amphibian material exposed to calf thymus RNA, though Yamada (1961), using identical p...
